In Vivo Osteogenic Potential of cultured Cell Derived from the Chondrocyte

Abstract

Chondrocyte were isolated from costochondral cartilage of young rabbits, introduced into cell culture, subcultured and then inoculated into abdominal wall of adult rabbits to test the vivo osteogenic potential of cultured chondrocyte. In
monolayer
culture, the adherent chondrocyte showed fibroblast-like morphology and expressed neither osteogenic nor chondrogenic phenotypes. Cultured cells inoculated cells inoculated hetertopically into adult rabbit showed only chondrogenic eviddence with
formation of chondrocyte nest at the subcutanous injection site.
These observation provided the base of exploring the combination of such cultured chondrocyte with porous hydroxyapatite and implantation heterotopically in adult rabbit to test the osteochondrogenic potential of this composite graft These cells
eventualiy gave rise to bone tissue in the pore of hydroxyapatite at the heterotopic implantation site
These results raise the possibility that composite graft of cultured chondrocyte-derived cells and porous hydroxyapatite can be clinically used as a bone graft substitute in situation requiring bone augmentation or regeneration.
